Congratulations to Prof Jamilur Rahman, professor of Department of Genetics and Plant Breeding at Sher-e-Bangla Agricultural University and his collaborators at Max Planck Institute for Chemical Ecology, Germany for their breakthrough research on Viral Induced Gene Silencing system. . In the research paper, published in BMC plant biology, they have reported a novel Virus-Induced Gene Silencing (VIGS) system to study the functional analysis of plant gene(s) at elevated temperature conditions. Typically, VIGS method requires growth temperature lower than those of the plant's native environment. Development of this enhanced thermal tolerant VIGS vector system is very promising in terms of ecological research. The research group have developed this system based on a Tobacco Rattle Virus (TRV) California isolate. The viral genome was used to construct separate binary plant transformation vectors for functional elements of RNA1 (6765 nt) and RNA2 (3682 t). Transient transformation with both vectors in the host plant was used to induce VIGS of target genes. Treating the host plant with sap from already VIGS induced plants was also used as an alternative method. They observed the silencing efficiency of PDS ( Phytoene desaturase) gene in Nicotiana attenuata in different temperature. Silencing efficiency at 28 °C and 30 °C was 90% and 78% respectively. This new TRV VIGS vectors induce gene silencing in Nicotiana attenuata at higher temperatures than the existing pBINTRA6/pTV00 vector system which can be due to the differences in the suppressor protein and RNA dependent RNA polymerase sequences. Though it can cause greater growth defects but it opens up avenue to study gene function even in field condition due to their elevated thermal stability.
